Using a lab-scale activated sludge system equipped with an external selector for suppressing nitrite oxidising organism (NOO), the microbial population dynamics were investigated during about 400 days of the continuous operation. In the varied selector operations and excess sludge withdrawal, the nitrification, partial nitritation, and nitrification loss followed by the recovery of partial nitritation consecutively took place. The DNA analysis with the high-throughput sequencing revealed that only Nitrosomonas spp. retained during the nitritation–nitrification and only Nitrospira spp. could grow in nitrification, respectively. Proteobacteria was the most predominant heterotroph in the main aeration tank, whilst Bacteroidota was abundant in the external selector in association with the reduced microbial diversities. In the external selector, due to the high nitrite and ammonium, the ammonium oxidising organism’s (AOO) inherent enzyme genes (Amo and Hao) per total microbial gene copies were as low as about 50% of that in the main aeration tank. The dynamically calculated AOO and NOO concentrations using the IWA-ASM1-based model could be linearly correlated to both relative abundances in the activated sludge. The correlations suggested that the DNA analysis could be potentially utilised as an alternative tool to estimate the nitrifier biomass of activated sludge instead of performing kinetic modelling.

In this study, we examined the anaerobic incubation of waste-activated sludge (WAS) under thermophilic conditions. First, we reconfirmed CO2 sequestration using WAS without any enrichment process at different temperatures under mixed gases (CO2:H2 = 1:4). We observed that the methanogens, which were previously enriched at 37°C, displayed strong CO2 sequestration at an early incubation time. Although CO2 sequestration at 60°C showed the largest impact after 6 days, no methane production was observed at each thermophilic condition within 4 days. Therefore, we enriched methanogens using a novel WAS protocol for 10 days under nitrogen gas under thermophilic conditions. These methanogens displayed better CO2 sequestration than the original strain at 37°C. Moreover, methane production by the thermophilic methanogens was due to the hydrogenotrophic pathway rather than the acetoclastic one because low methanogenic activity from acetate was observed. Microbial community structure analysis showed that the Methanothermobacter genus belonging to hydrogenotrophic methanogens was predominant among the thermophilic enriched methanogens at 60°C. Taken together, the thermophilic methanogens can be a useful tool to sequestrate CO2 efficiently.

With the recent increase in demand for biodegradable plastics triggered by the microplastics problem, the reuse of waste is also attracting attention. For example, VTT Technical Research Centre of Finland holds a patent to produce high-purity oligo(lactic acid) (OLA) from polylactic acid waste. If a new useful strain capable of assimilating the OLA can be discovered, it will be possible to produce useful substances from polylactic acid-based waste. For that purpose, it is necessary to clarify the relationship between bacteria and the OLA. As part of this, we tried to analyze the hydrolysis behavior of oligo(lactic acid) using reverse-phase HPLC. From the results, it was shown that the addition of trifluoroacetic acid (TFA) to the HPLC eluent significantly suppressed the broadening of the peaks derived from the OLA, enabling the quantification of OLA. The analytical method will be useful for investigating the effects of useful bacteria on the OLA.

Nucleic acid extracted from environmental samples is an important analyte in instrumental analysis. With respect to the phenomenon in which sodium tungstate promotes the methane production in anaerobic digestion, the analyses of microbial community using RNA were conducted; however, we noticed that only RNA concentration measured by absorbance measurement (NanoDrop) was remarkably high. Therefore, in this study, DNA and RNA were extracted from the anaerobic digestion sludge samples with sodium tungstate, sodium selenite, or sodium molybdate, and these nucleic acids were quantified and compared by absorbance measurement, fluorescence measurement (Qubit), and gel electrophoresis. Interestingly, it was found that only the RNA concentration of the sample containing sodium tungstate measured by NanoDrop was 3 times higher than that by Qubit analysis. In addition, there was no difference between the RNA concentration measured by Qubit and gel electrophoresis. Regarding DNA concentration and the other compounds, there were no differences. Hence, these results indicate that the Qubit system is useful for the quantification of the RNA concentration in the environmental samples.

Most palm oil mills adopted conventional ponding system, including anaerobic, aerobic, facultative and algae ponds, for the treatment of palm oil mill effluent (POME). Only a few mills installed a bio-polishing plant to treat POME further before its final discharge. The present study aims to determine the quality and toxicity levels of POME final discharge from three different mills by using conventional chemical analyses and fish (Danio rerio) embryo toxicity (FET) test. The effluent derived from mill A which installed with a bio-polishing plant had lower values of BOD, COD and TSS at 45 mg/L, 104 mg/L, and 27 mg/L, respectively. Only mill A nearly met the industrial effluent discharge standard for BOD. In FET test, effluent from mill A recorded low lethality and most of the embryos were malformed after hatching (half-maximal effective concentration (EC50) = 20%). The highest toxicity was observed from the effluent of mill B and all embryos were coagulated after 24 h in samples greater than 75% of effluent (38% of half-maximal lethal concentration (LC50) at 96 h). The embryos in the effluent from mill C recorded high mortality after hatching, and the survivors were malformed after 96 h exposure (LC50 = 26%). Elemental analysis of POME final discharge samples showed Cu, Zn, and Fe concentrations were in the range of 0.10–0.32 mg/L, 0.01–0.99 mg/L, and 0.94–4.54 mg/L, respectively and all values were below the effluent permissible discharge limits. However, the present study found these metals inhibited D. rerio embryonic development at 0.12 mg/L of Cu, and 4.9 mg/L of Fe for 96 h-EC50. The present study found that bio-polishing plant installed in mill A effectively removing pollutants especially BOD and the FET test was a useful method to monitor quality and toxicity of the POME final discharge samples.

This study investigated the fate of Escherichia coli (E. coli) O157 spiked into 24 commercially available fermented soybean paste (miso) samples. Viable cell numbers of the inoculated E. coli O157 at storage conditions of 5, 10, 20, or 30°C were measured by the agar plate method or most probable number method. Cell numbers of E. coli O157 decreased gradually in all tested samples. The decreases in E. coli O157 numbers depended mainly on the water activity of samples. Decreases in the contamination level of E. coli O157 did not depend on the ingredients of miso samples (malted rice, wheat, and soybean). The obtained results suggest that the commercial risk of accidental E. coli O157 contamination is practically negligible because of high cell mortality during transportation and storage processes at room temperature.
