    <p>A new era of life science research has arrived in the use of electron cryo-microscopy. The combination of electron cryo-microscopy with single-particle analysis enables us to analyze structures of proteins and their complexes at the atomic resolution, which is becoming a key technology of structural biology. In addition, the combination of cryo-EM and electron tomography is revealing new and marvelous views of cells and tissues. However, cryo-EM images of living organisms and their components have a low signal-to-noise ratio, due to weakness against electron dosages, in terms of information technology, and thus automatic image acquisition system is necessary because a large number of images are required. In this paper, we will introduce the current efforts of automatic electron microscopy for this purpose, including the system we have been developing. From the standpoint of ‘System of Systems’, we will discuss open system and IoT as specifications for the automation of electron microscopy systems, and propose the necessity of a feedback system based on procedure logs.</p>

    <p>Cryo-EM single particle analysis can solve multiple protein structures contained in a sample by classification. However, the information on the dynamics between the solved structures is lost and it can only be inferred. About this problem, cryoDRGN, a deep learning approach for solving the three-dimensional reconstruction and structural classification that was announced in 2020, breaks away from discrete data partition. The method is based on an auto-encoder, and realizes continuous structural classification by constructing a latent space that separates the information depending on the projection parameters from the input particle image. In this paper, we explain conventional classification method in single particle analysis and deep learning topics that are the background of cryoDRGN. Then, as a benchmark for structural classification, we try three-dimensional reconstruction on the actual data of GroEL/ES having 6 kinds of complexes.</p>

    <p>The molecular mechanism through which how beta decays in tritium-substituted species damage DNA and polymeric materials is still unknown. Molecular dynamics simulations of hydrogen-removed polyethylene were performed to predict the structural change of the polyethylene chain after the substituted tritium decays. We calculated the potential energy, the global orientational order parameter, and the average number of consecutive <i>trans </i>bonds. The results are that, the greater the number of removed hydrogen atoms, the higher the potential energy and the lower the value of the global orientational order parameter and the average number of consecutive <i>trans </i>bonds. Thus, after losing hydrogen, polyethylene becomes poorer in terms of both thermal and structural stabilities.</p>

    <p>The electron cryo-microscopy is one of the most powerful techniques to elucidate biological molecular structure under nearly physiological condition. By single particle analysis, many three-dimensional structures of proteins and their complexes have been resolved at atomic resolutions. Furthermore, <i>in situ</i> molecular behaviours in cells and tissues have also been reported at nano-meter or sub-nanometer resolutions by electron tomography. I would here review current situations of electron cryo-microscopy and the principles of their analysis and so show its possibility.</p>

    <p>We propose a computational strategy for investigating structural change of tritium-substituted macromolecules. Effects of radiation on macromolecules such as polymeric materials and DNA are classified into three categories: (1) direct action, (2) indirect action, and (3) decay effect. In this study, we focus on the decay effect exclusively. After a beta decay of substituted tritium in macromolecules to helium-3, the generated inert helium-3 is assumed to be deleted quickly. To get an insight into the decay effect to the damage of macromolecules, we perform molecular dynamics simulations of tritium-deleted macromolecules and analyze their structural change. Preliminary simulation results of decay effect on polymeric materials and DNA are presented.</p>

    <p>The cryo-electron microscopy is one of the most powerful techniques to elucidate biological molecular structure under nearly physiological conditions. Furthermore, we can also visualise in situ molecular behaviours in cells and tissues at nano-meter resolutions by electron tomography. We here clarified the architecture of filopodia, which is an essential device for cell motility and sensing. We observed the hexagonal actin bundles and their sets, where the actin numbers in the bundles are consistent with those estimated by mechanical balance. The cross-linked structures between actin filaments were averaged and compared with the atomic model resolved by X-ray crystallography; we showed a binding manner of fascin to actin filaments. Besides, we browsed in reconstructed filopodia and so found cooperative bindings of fascin and short actins in the periphery of the actin bundles. Thus we proposed a novel filopodia formation mechanism. We challenged the elucidation of the elongation mechanism by observing the tips of filopodia. Hereafter, cryo-electron tomography will useful for materials in water as well as biological samples.</p>

    <p>We repot on the hybrid fine particles using core silica particles adsorbed on the ultra fine nano particles such as nano silica particles (ca.4nm diameter) and fullerenol for Chemical Mechanical Polishing of Hard to process materials. In our study, chemical reactivity and mechanical action from the hybrid particles were investigated using the several diameters of the core silica particles. At the case of adsorption of the nano silica particles and fullerenol, chemical reactivity become dominant parameter under the small size region although the nano particle is coated on the surface of the core silica particles. These results expect that both parameters as chemical reactivity and mechanical action can be control by the sizes of core particles and coating condition of the nano silica particles. We also optimize the structure of the hybrid silica particles at the case of the nano silica particles and/or fullerenol to meet the high performance CMP of hard to process materials and discuss about these results.</p>

    1,2-Dilauroyl-<i>sn</i>-glycero-3-phosphocholine (DLPC) is one of the major lipids constituting cellular membranes. When DLPC itself is dispersed in aqueous solutions, multilamellar vesicles are formed, although the cellular membranes are made of single bilayers. We found that the addition of a calix[4]arene derivative (CaL[4]C3) bearing four amine groups as the headgroup to the DLPC multilamellar vesicle induced the structural transformation to a single-lamellar vesicle, by use of small-angle X-ray scattering and electron microscopy, indicating that the highly condensed ionic groups of CaL[4]C3 are useful to control membrane morphology.

    In this study, we expressed, purified and characterized small heat shock proteins (sHsp) from a magnetotactic bacterium, <i>Magnetospirillum magneticum</i> AMB-1, Hsp17.5, Hsp17.8 and Hsp18.3. They all showed molecular chaperone activity to protect citrate synthase from thermal aggregation at 45°C. The oligomeric states were examined by size exclusion chromatography and electron microscopy. Hsp17.8 exists as a spherical oligomer like other sHsps, and the oligomer reversibly dissociates to small conformers, probably dimers, at elevated temperatures. On the contrary, Hsp17.5 and Hsp18.3 take large filamentous conformations. A similar structure was observed in an sHsp of <i>Sulfolobus tokodaii</i>, StHsp19.7, but it did not exhibit molecular chaperone activity. Hsp17.5 and Hsp18.3 changed their oligomeric states according to the elevation of temperature. Among them, Hsp17.5 exhibited reversible dissociation. This is the first report that the filamentous sHsp exhibit molecular chaperone activity with the change of oligomeric states.<br>
