We developed a novel technique for measuring neuronal extracellular potentials in a neuron-astrocyte co-culture system using a SiN (silicon nitride) membrane having microelectrodes and microholes. An 8×8 microelectrode array was formed on one side of a 1 μm-thick SiN membrane, and multiple microholes of 5 μm in diameter were opened through the membrane. Then, neurons were cultured on the microelectrodes after astrocytes were cultured on the back side of the membrane. The neuronal spikes measured in this novel back-to-back co-culture method were compared with those in the conventional method in which both of neurons and astrocytes were co-cultured on the microelectrodes. In the conventional method, formation and enlargement of cellular aggregates made neurons out of touch with microelectrodes, and posed measurement of a small number of spikes. On the other hand, our novel method permitted the firm contact between neurons and microelectrodes and the cell-to-cell interaction through microholes, and therefore neuronal spikes increased drastically with culture time, which led to stable measurement of neuronal extracellular potentials.

To measure the electrical signals of astrocytes, we developed a back-to-back (BTB) layered co-culture device which has a microporous SiN (silicon nitride) membrane equipped with a microelectrode array (MEA). Astrocytes were cultured on the electrode-formed surface of the membrane, and neurons were also cultured on the back surface. Then, the signals detected by the device were compared with those detected by a conventional MEA device where neurons and astrocytes were co-cultured on the same surface having microelectrodes. As a result, we demonstrated that the BTB layered co-culture device can isolate the astrocyte signals from a neuron-astrocyte co-culture system.

We developed a novel microelectrode array device for multichannel measurement of extracellular potentials of neurons which are co-cultured with astrocytes. 64 microelectrodes were formed in an 8×8 matrix on a SiN (silicon nitride) membrane of 2 μm in thickness having multiple microholes of 6 μm in diameter. Then, neurons and astrocytes were back-to-back co-cultured on both surfaces of the membrane. We succeeded in recording electrical signals caused by spontaneous neuronal activity on the 9th day after the beginning of co-culturing.

We developed a technique for co-culturing neurons and astrocytes on both sides of a 1-µm-thick SiN membrane which has multiple ordered microholes measuring 3 μm in diameter. Astrocytes were cultured on a membrane surface which was coated with collagen, and neurons were cultured on the other surface which was coated with laminin. We prepared three samples having different astrocyte densities, and counted the number of fluorescently stained synapses in order to evaluate neuronal activity. As a result, the number of synapses increased with an increase of astrocyte densities, and also increased with co-culture time.

© MYU K.K. Positioning and patterning of polystyrene particles on a silicon nitride (SiN) membrane with a microhole array has been demonstrated by positive dielectrophoresis (p-DEP). A chamber with an SiN membrane with the well-aligned microholes as the bottom substrate was positioned on an indium-tin-oxide (ITO) electrode with a 1 mm space between the bottom substrate and the ITO electrode. The chamber and the space were filled with water and a suspension of particles (10 µm diameter) in water, respectively. An AC electric signal was then applied to a microelectrode positioned at 10 µm above the SiN membrane, while the ITO electrode was connected to the ground. Particles present in the space between the SiN membrane and ITO electrode gradually moved toward the lower surface of the SiN membrane directly under the microelectrode owing to the strong electric field generated on and in localized microholes and accumulated at this position to form aggregates. For particles of 3 µm diameter, one particle was deposited in each hole (2 µm diameter) in the region directly under the microelectrode. The horizontal movement of the microelectrode gave rise to the formation of a line pattern of particles along the trail of the microelectrode because of the shift of the region with the strong electric field. These demonstrations could be applicable to arranging the particles at desired positions and in desired holes, and form particle patterns with highly flexible designs.

We developed a technique for producing and separating micrometer-size vesicles from human lymphocytes using microholes. Using cell-anchoring molecules, lymphocytes were immobilized on a microhole array formed in a silicon nitride (SiN) membrane. While the cells were chemically stimulated with sodium butyrate (NaB), vesicles were produced inside the microholes and passed through them. We tested microholes measuring 2, 4, and 5.5 μm in diameter, and counted the number of vesicles and cells which passed through microholes. In case of microholes 4 μm in diameter, a large number of vesicles were obtained, and the cell invasion rate was below 0.5 %.

For long-term culture of a single neuron, many astrocytes and a single neuron were co-cultured on both sides of a 1 μm thick silicon nitride membrane which has many microholes of 3 μm in diameter. After astrocytes were cultured on the back side of the membrane for 1 week, a neuron was seeded on the front side of the membrane of a microwell bottom and cultured for 5 weeks. By fluorescently staining VGLUT1 (vesicular glutamate transporter 1) which is localized at a presynaptic terminal, we found that the number of synapses formed by a single neuron drastically increases within the first 2 weeks and keeps high values for weeks afterward.
